The instruction sheet I believe tells you what the stock setting is for the 2nd gear. Too, are you for certain that you are turning the right set screw? It is likely that you could be taking off in 2nd, but the best way to tell is throttle up slowly and just listen for the drop of rpms on the engine. If it just continues to rev up, but doesn't appear to be moving very fast then it's just not going into 2nd, but if it revving up and moving pretty fast then your are right you are stuck in 2nd. Either way 1st gear should engage, even if only for a second on take off. Do that simple test and see where you get after setting it back to the stock setting. I don't have my manual so I really can't tell you what the setting is, sorry.

Hi there, turn the screw clockwise all the way closed, then turn it anti-clockwise for 4.5 turns, that is the factory setting for the Savage .25, if it needs adjustment, turn a quarter at a time, clockwise will give a later shift, and anti-clockwise will give an earlier shift

RE: adj 2 speed on 25
I just checked my Savage update sheet that came with my 3 speed LE and it lists the standard positions for the shift screws
as :
using the 15t / 49t 3rd gear 3 1/2 turns "external adjustment", 2nd gear 3 1/4 turns "adjustment inside trans".
using the 19t / 47t 3rd gear 5 turns, 2nd gear 4 turns
The way the manual indicates is that this is additional turns CW from having the set screw flush at the top of the hole for 2nd and
even with the lower edge of the hole for 3rd not from a fully tightened position.

As far as turns out from turned in , you could count the turns from that position and compare it to the spec,
along with test driving / adjusting.
My truck has been shifting well and I have not changed mine yet so I do not know for sure, I can only reference the manual.
